TY - JOUR AB - The grey short-tailed opossum, Monodelphis domestica, has been an established research animal for more than five decades, but relatively, little is known about its thermophysiology. Here we studied core body temperature (T b) and metabolic rate (MR) of female adult M. domestica housed in the laboratory at an ambient temperature (T a) of 26 °C. In expanding previous reports, the average recorded core T b of M. domestica was 34.3 °C. The T b of an individual M. domestica can drop below 30 °C (minimal T b: 28.6 °C) accompanied by a reduction in MR of up to 52 % even while having ad libitum access to food. These findings demonstrate for the first time the presence of spontaneous torpor in M. domestica. Metabolic suppression at relatively high T a and T b furthermore broadens our perspective on the use of torpor as a metabolic strategy not just restricted to cold climates. Phenotyping or phenotypic screening involves the comparison of wild-type control mice with their mutant or transgenic littermates. In our study, we explored the extent to which standardized phenotyping will succeed in detecting biologically relevant phenotypic differences in mice generated and provided by different collaborators. We analyzed quantitative metabolic data (body mass, energy intake, and energy metabolized) collected at the GMC under the current workflow, and used them for statistical power considerations. Our results demonstrate that there is substantial variability in these parameters among lines of wild-type C57BL/6 (B6) mice from different sources. Given this variable background noise in mice that serve as controls, subtle phenotypes in mutant or transgenic littermates may be overlooked. Furthermore, a phenotype observed in one cohort of a mutant line may not be reproducible (to the same extent) in mice coming from a different environment or supplier. In the light of these constraints, we encourage researchers to incorporate information on intrastrain variability into future study planning, or to perform advanced hierarchical analyses. The mean erosion and deposition rates derived from Cs were -37 and +52 t·ha-1·year-1, respectively; those from Pu were -32 and +39 t·ha-1·year-1. We found no statistical difference between the means obtained by the two tracers. In contrast to Pu, however, the rates obtained by Cs were not accurate enough to assure the presence of a net soil redistribution. Based on experiments on mice using high doses, the genetic risk for humans has to be derived by extrapolation to low doses, assuming mouse and man respond in a similar way. Microdosimetric considerations indicate a linear relationship; studies on the molecular mechanism of radiation-induced mutagenesis lead to the conclusion that, in addition to a constitutive mutagenic process, an inducible compound exists which may cause linear-quadratic dose-effect curves. Strains 11 X and RH 12 are characterized by the bending type of cell division, a GC content of 70.2 and 70.5%, ll-diaminopimelic acid in the cell wall, absence of metachromatic granules, utilization of several sugars and no changes in cell morphology by citrate.
