The role of FGFR2 in cerebellum development and depressive disorders.

The main goal of this study was to analyse genetic factors and pathway involved in depressive disorders using mice as animal models. I inactivated FGFR2 in the entire CNS from day 11.5 of embryonic development in mice and my histological analysis revealed cerebellar malformations. I could show that these cerebellar phenotypes are due to developmental defects, probably caused by abnormal Bergmann glia development and morphology. Furthermore, I showed that the restricted inactivation of FGFR2 in forebrain neurons in adult mice altered depression-like behaviour and the acute response to the antidepressant fluoxetine in the forced swim test. Thus, FGFR2 could play a causative role in depressive disorders and mediate behavioural effects of antidepressants. Additionally, I applied the RNAi technique to generate P2RX7 “knockdown” mice as a new model for Depression.
Das Ziel der Doktorarbeit war die Analyse genetischer Faktoren bei depressiven Erkrankungen. Ich habe FGFR2 im zentralen Nervensystem (ZNS) von Tag E11.5 an im Maus-Embryo inaktiviert. Meine histologischen Analysen zeigten eine Kleinhirnfehlbildung, die wahrscheinlich durch gestörte Bergmann-Glia-Entwicklung und –Morphologie verursacht wird. Die eingeschränkte FGFR2-Inaktivierung in Vorderhirnneuronen von erwachsenen Mäusen führte zur Veränderung sowohl des depressionsbezogenen Verhaltens als auch der akuten Reaktion auf das häufig verwendete Antidepressivum Fluoxetin. Daher scheint FGFR2 ursächlich an depressiven Erkrankungen und an der Wirkung von Antidepressiva beteiligt zu sein. Ferner habe ich mittels RNAi-Technologie eine Mausmutante für P2RX7 als weiteres Mausmodell für Depression hergestellt.
