The highly conserved transactivation response DNA binding protein TDP-43 represents the major pathological component in inclusions of (moto-) neurons in FTLD and ALS. Homozygous mice of TDP-43 null mutants showed an embryonic lethal phenotype and died within the first week after fertilization. A tetraploid rescue showed an extraembryonic contribution to the mutant embryonic phenotype. Heterozygous individuals showed an upregulation of TDP-43 to 70-80%, indicating a feedback loop regulating the expression of TDP-43 on the mRNA level.
