Animal experimentation with radium 224.

The examinations on early damage on the skeleton were carried out in the proximal fibia metaphysis of rats and mice after incorporation of 224Ra. In addition to the conventional histological examinations, enzyme-histochemical, electrone-microscopic, and autoradiographical methods of examination were applied. The results are briefly shown. The examinations concerning the question of delayed damage were mainly carried out on female NMRI-mice which had been administered intraperitoneally 1.cal, electrone-microscopic, and autoradiographical methods of examination were applied. According to this, the osteosarcome incidency was between 7 and 20% without clear dose dependency. The skeletal distribution of osteosarcomas, however, was dose-dependent. In the experiments with repeated injection of 224Ra, an increase in the multifocal osteosarcomas could be observed. The results of these experiments presented here cannot be directly applied to men.
Die Untersuchungen ueber die Fruehschaden am Skelett wurden im der proximalen Tibiametaphyse von Ratten und Maeusen nach Inkorporation von 224Ra durchgefuehrt. Ausser den konventionellen histologischen Untersuchungen wurden enzymhistochemische, elektronenmikroskopische und autoradiographische Untersuchungsmethoden herangezogen. Die Ergebnisse werden kurz vorgestellt. Die Untersuchungen zur Frage der Spaetschaeden wurden vorwiegend an weiblichen NMRI-Maeusen durchgefuehrt, denen intraperitoneal 1,0 - 50 μCi/kg 224Ra injiziert wurden. Die Osteosarkominzidenz lag danach zwischen 7 und 20% ohne eine klare Dosisabhaengigkeit. Die Skelettverteilung der Osteosarkome dagegen war dosisabhaengig. In den Experimenten mit mehrmaliger Injektion von 224Ra wurde eine Zunahme von multifokalen Osteosarkomen beobachtet. Die vorliegenden Ergebnisse koennen aber nicht ohne weiteres auf den Menschen uebertragen werden.
