Specific Locus Test.

The mouse specific-locus mutation test is the most efficient method to screen for de novo spontaneous or induced recessive mutations in the germ line of a mammalian species. The loci screened, the experimental protocol, and the historical background to the development of the specific-locus test are described. Not only have results from specific-locus mutation experiments provided most of the experimental data on the mechanisms and rates of mutations occurring in mammalian germ cells following radiation or chemical mutagen exposure, but the mutations recovered from such experiments have also been a valuable source of genetic variation in the mouse for subsequent genetic and molecular studies.
