Microbiome-Based Therapies: Phage Intervention.
In: The Gut Microbiome in Health and Disease. 2026. 525-552 (Gut Microbiome in Health and Disease A Textbook)
The human microbiome is linked to various health benefits and disorders. As part of the human microbial community, bacteriophages (phages) play a crucial role in regulating the microbial communities within the human gut, concomitantly giving rise to a novel research field: the human virome. Importantly, as these phages are viruses that target bacteria in a host-specific manner, various phage-based therapeutics could be developed for treating gut disorders. In this chapter, different aspects of phage therapy as a tool for modification of gut microbial composition and functionality are discussed within the context of various intestinal diseases. The discussions include the use of fecal virome transplant as a delivery method, the application of phage-derived enzymes as antibacterial agents and the available in vitro and in vivo models for testing phage therapy. Challenges related to phage therapy are also assessed, ranging from the development of phage-resistant bacteria, non-optimal phage cocktails, and inactivation of phage particles in the human body.
